一微生物簡介
微生物:是一類肉眼看不見,有一定形態(tài)結構,能在適宜環(huán)境中生長繁殖的細小生物的總稱。
微生物也像動物、植物一樣,具有生命,它可以由小長大,可以“生兒育女”,繁殖后代,也可以“吃”進食物,排出廢物,它也會死亡。也就是說生命體(無論動物、植物)所有特征它都具有,只不過是它的個子特別特別的小,小到我們根本無法用肉眼看到它。只能借助顯微鏡才能看到它。當然,有的大型微生物我們用肉眼也可以看到,如蘑菇、木耳等,盡管它們比較大,但我們也把它歸于微生物這一類。微生物這個家庭非常龐大,成員也很多,根據(jù)它們的一些特點,我們把它分成細菌、放線菌、真菌和病毒四大類。
二培養(yǎng)基
培養(yǎng)基是供微生物生長、繁殖、代謝的混合養(yǎng)料。由于微生物具有不同的營養(yǎng)類型,對營養(yǎng)物質(zhì)的要求也各不相同,加之實驗和研究的目的不同,所以培養(yǎng)基的種類很多,使用的原料也各有差異,但從營養(yǎng)角度分析,培養(yǎng)基中一般含有微生物所必需的碳源、氮源、無機鹽、生長素以及水分等。另外,培養(yǎng)基還應具有適宜的pH值、一定的緩沖能力、一定的氧化還原電位及合適的滲透壓。 瓊脂是從石花菜等海藻中提取的膠體物質(zhì),是應用最廣的凝固劑。加瓊脂制成的培養(yǎng)基在98~100℃下融化,于45℃以下凝固。但多次反復融化,其凝固性降低。 任何一種培養(yǎng)基一經(jīng)制成就應及時徹底滅菌,以備純培養(yǎng)用。一般培養(yǎng)基的滅菌采用高壓蒸汽滅菌
